You can take a ferry to reach there by landing on Outer Harbour Ferry Terminal, where you can take the free shuttle bus of Venetian to the hotel resort and have a simple glance if you have interest.

Hereunder is an itinerary for one-day M trip, through which you can have an overall knowledge of this city:

Ruins of St. Paul - Largo do Senado (Senado Square) - Rua do Dr. Soares - Rua Central - Avenida da Praia Grande - Avenida da Boavista - Calcada da Penha - Estrada de D. Joo Paulino - Rua de S. Tiago da Barra - A-Ma Temple - Fisherman Wharf

As they are all within walking distance from each other, you can enjoy a hiking travel:

Upon your getting off from the free shuttle bus, you can walk to the ruins.
Start from the ruins. You can walk to Largo do Senado after visiting.
Then turn left on to Calcada do Tronco Velho, along which you can walk to Largo do Santo Agostinho. It is paved in cobblestones and lined with antique green streetlights.
On the left side of that is a famous church - Igreja de So Agostinho in Portuguese.
After that, walk along Rua do Dr. Soares, Rua Central, Avenida da Praia Grande, Avenida da Boavista, Calcada da Penha, Estrada de D. Joo Paulino and Rua de S. Tiago da Barra, and finally reach the A-Ma Temple.
At last, if you have time left, you can take a bus to Fisherman Wharf in 15-20min.
May help and good day!

It is a walking distance from fisherman's wharf to the Marine Ferry Terminal. From the tower, you can take bus 32 to the ferry terminal first, then walk to the Fisherman's Wharf. After visiting it, walk back to the ferry terminal to ferry to Hong Kong.
