Amberly, you will have to exit mainland China toward Macau or Hong Kong and take your flight from one of those two territories toward Singapore. The basic rule of 240 hour visa free transit requires that the immediate points before and after mainland China are located in two different countries or eligible territories (eligible territories are Hong Kong, Taiwan and Macau). In other words, you cannot have an itinerary: Singapore - Guangzhou - Shenzhen - Singapore because both immediate points are in just one country - Singapore. If you insert Macau or Hong Kong, your itinerary will look like: Singapore - Guangzhou - Shenzhen - Macau (or Hong Kong) - Singapore. This is definitely eligible for 240 hour visa free transit because the immediate points will be located in two different countries / territories, Singapore and Macau (or Hong Kong). It is important to understand that you will have to buy your ferry ticket from Shekou (a port in Shenzhen) to Macau or a train ticket from Shenzhen to Hong Kong in advance, even before you depart from Singapore because you will be asked to present one in Singapore (at the time of your check-in) and in Guangzhou (upon your landing there).
