Emelie, the first thing first. You did not mention the way how do you think to reach Guangzhou from Hong Kong.
You cannot do it in any way if you want to enjoy 144 hour visa free transit scheme, but so far the only way is by air on a direct flight from Hong Kong to Guangzhou or by ferry from Hong Kong to Nansha port located south from Guangzhou.
Also, you cannot return from Guangzhou via Hong Kong because of the simple rule that clearly says that the immediate points before and after your Chinese mainland destination have to be in two different countries or eligible territories (eligible territories are Hong Kong, Taiwan and Macau).
In other words, you can enter Guangdong province from Hong Kong in the way I explained, but you have to exit to a third country or eligible territory. If you ask me, I would suggest you to exit Guangdong province to Macau by ferry from Shekou port in Shenzhen.
So, your itinerary would be:
Hong Kong - Guangdong province (where Guangzhou, Shenzhen and many other cities are) - Macau.
Everything before and after this segment is not of interest to Chinese immigration authorities. You should be aware that you will have to buy your ferry ticket from Shekou port in Shenzhen to Macau even before you depart from Hong Kong to Guangzhou as a proof that you will be really in a transit. Also, you will print it out and have at hand ready to be presented to relevant authorities. In the end, you can adapt your flight itinerary to fly from Guangzhou to some other country/territory different than Hong Kong.
