As your visa has only one entry, you can only enter to Mainland China for once. If you only transit in Shanghai and stay for no more than 24 hours in Shanghai then fly to HK, you are eligible for the 24h visa free transit, which won't waste your single entry, and then enter Shenzhen from HK you can use the entry.
But if you stay in Shanghai for over 24 hours, you will have to use the single entry, and when entering Shenzhen, you need to apply for the Shenzhen visa on arrival at Huanggang, Luohu or Shekou port, and you may fail to get it; or apply for another visa in HK.
