You can do that in three ways:

1. You can apply for a 5 day Shenzhen visa on arrival at one of three designated border crossings between Hong Kong and mainland China. The price of this visa is 130 CNY, but you have to understand that you are limited to Shenzhen city limits only. The most convenient border crossing is Shekou that you can reach by taking the first morning ferry from Hong Kong. After your stay in Shenzhen, you can return back to Hong Kong by taking any border crossing and any mean of transport, high speed train included.
2. You can apply for a usual single entry Chinese visa at Lo Wu (Luohu) border crossing that is valid for 30 days and that allows you to travel within the entire China in the way as you wish. The price of this visa for US citizens is 971 CNY.
3. You can rely on 144 hour visa free transit, but in this case you will have to do the following:
- Fly from Hong Kong to Guangzhou
- Buy your ferry ticket from Shekou port in Shenzhen to Macau even before you depart for Guangzhou and have that ticket printed out and at hand and you will also have to take exactly that ferry you have bought your ticket for.
- You will exit mainland China for Macau
- Like this you will be able to stay within the entire Guangdong province for up to 144 hours.

Hope this helps :)
