About:240-Hour Visa-Free Transit

US- Tokyo, Japan (layover)- Shanghai- US

Does this itinerary eligible for the 144-hour-visa-free transit? Or should I stay in Tokyo for some time before I fly out to Shanghai?

There's no requirement to stay in Japan for a minimum time. You don't even need to clear immigration.

But I do need to stay in Shanghai for 2-3 days. So can I do the 144hr transit?

Of course. Just make sure your Shanghai to USA flight does not touch down in Japan or mainland China.
