About:144-Hour Visa-Free Transit

Layover. Flying from US to Shanghai but layover in Tokyo (Japan)

then from Shanghai to Osaka (Japan) does it require VISA?

Yes you need a visa as you're not in transit.
You fly Japan - China - Japan and that's not a transit.

Even if I originate from the US? I don’t even leave the airport during layover. The layover is only 3hrs

Only the stop immediately before Shanghai matters and not your point of origin. You can elect to apply for a visa for $140 or change your ticket, otherwise boarding/entry will be swiftly refused.
