About:72-Hour Visa-Free Transit

transit 5 hours in Shanghai airport from Bangkok to New York.

If we can use 72 hours free option; do we have to claim our luggage or just leave/entry airport and claim it at final destination?

For 5 hours transit you will be granted 24 hours direct transit, not 72 hours. You should ask your airline if they will check your luggage to your final destination.
