Can I book a return plane flight from Hong Kong to Ganngzhou

and then take advantage of the 72 hour visa free visit

You mean you will fly to Guangzhou from HK and then fly back to Australia? If yes and your stay in Guangzhou is less than 3 days, you are eligible for the 72 hour free transit in Guangzhou.
