About:Guangzhou Baiyun International Airport

I have a layover in Guangzhou from 2:25 pm to 1:50 am and am traveling from Vietnam to USA.

Do I need to apply for a 72 hour visa? Also, would I need to collect my luggage during the layover? If so, where could I leave my luggage and pick it back up late at night (approximately 11:50 pm)?

Oh, theoretically, as long as you are from the country which is among the 51 countries eligible for the policy, you can enjoy it. However, your layover is within 24h, so you can use the 24h visa-free transit, for which you don't need to collect your luggage. Just show your luggage tap receipt and the staff will handle it to your final destination. If wishing to have a rest, you can go to the Pullman Airport Hotel opposite the terminal building.
