About:Shanghai Pudong Airport

Can I only fly to and from Shanghai for the 72 hour visa from Japan as a US citizen?

No, you cannot. This 72h visa free policy does not apply to passengers who travel on round trip flights. Your departing site and destination have to be different international areas.
