About:Shanghai Pudong Airport Shuttle Bus

We have a plane at 5.30 AM in Shanghai Pudong.

so we would need to take a bus around 2.30 AM to the airport. I see the night line goes from Pudong to the center but does it also go from the city center (Huashan Lu stop) to the airport around 2.30 AM?

No, at that time there is no reversing night line to the airport.
You may take a taxi or reserve a see-off service in advance.
