Hi, can I fly to Hong Kong (2 nights), then take a train to Shenzhen (2 nights) then to Guangzhou (2 nights) before flying back to Singapore.

You did not mention your citizenship. If you are a Singaporean citizen you do not need any kind of visa because Singaporean citizens are allowed to enter and stay in mainland China for up to 30 days visa free. If you are a citizen of a country that needs a visa to visit China, you will have to obtain one in advance.
