Your first itinerary is not eligible for any kind of visa free transit scheme because 144 hour visa free transit requires that only those arriving by plane into Guangdong province are eligible for this visa free transit scheme. You can, of course, fly from Hong Kong to Guangzhou, but I think that this option is not convenient for your needs.
Your second itinerary is eligible because you can exit Guangdong province toward Hong Kong or Macau without flying. Taking a train has become eligible way to exit Guangdong province, but this detail has not yet been loaded into Timatic. Consequently, until it is loaded, you are advised to take a ferry provided by CKS - Chu Kong Passenger Transport Company from either Nansha port south from Guangzhou or Shekou port in Shenzhen to Hong Kong. There is also eligible Humen port in Dongguan, but I do not see a way how to obtain a ticket for Humen - Hong Kong ferry in advance through Sea's Young website. So, you have to obtain a ferry ticket through CKS website in advance and print it out and hold it with you as a proof of your transit intention.
If you do as presented, you will be definitely eligible for 144 hour visa free transit and you do not need any kind of visa. Consequently, you should use correct terminology because this is not 144 hour visa, but visa free transit.
