Basically, the essence of this rule is that your immediate points before and after your mainland Chinese destination have to be in two different countries or eligible territories (Hong Kong, Taiwan and Macau). This is the reason why a flight from the immediate point to the mainland China airport has to be a nonstop one. In your case, Los Angeles does not play any role, but it is Manila that is your immediate point. Now, it is important that your flight Manila to Guangzhou is a nonstop type of flight.
As far as the ferry ports are concerned, both Shekou and Shenzhen Fuyong (so called Shenzhen airport support sea port) are eligible exits, so you can use both. I have to admit that I did not know about the closure of Shekou port during the last two weeks in May. Thank you for that. You will find three daily departures from Fuyong to Hong Kong provided by CKS.
Beside Fuyong port, you can also use Nansha port located south from Guangzhou if you find it more convenient, althoug Nansha offers two daily departures. Please, have in mind that you will have to book your ferry ticket in advance, print it out and have it with you. Also, you have to take exactly that ferry you have bought you advance ticket for.
Vaccination documentation is not needed. It is enough to do a DIY test and to scan/make a photo of its negative outcome. This test must not be older than 48 hours... It is quite possible that nobody will ask you about it. It is also required, although not exactly enforced that you complete China Customs’ Health Declaration Form. As far as your entry to Hong Kong is concerned, there are no specific requirements in this regard. You do not need anything regarding covid when you arrive in China.
