About:144-Hour Visa-Free Transit

Kuala Lumpur to Guangzhou by flight, then Guangzhou to HK by train, is it eligible?

Do you have to fly in and out? Or can you train out of Guangzhou to HKG?

One-way travel from Guangzhou to HK by train is permitted but only if you will be on the conventional Guangzhou-Kowloon through train and NOT the high-speed train. Airline and immigration will both need to see the ticket you booked in advance. For reverse you will have to fly HKG-CAN.
