Adriana, you have written your itinerary, but to be eligible for and kind of visa free transit scheme, you need also a city that will come after your Chinese destination that is, in your case, Guangzhou.

In other words, you have to construct your itinerary to look like:

Japan - Hong Kong - Guangzhou - X country (different than both mainland China and Hong Kong)

Also, you have to be aware that if you want to travel between Hong Kong and Guangzhou and to be eligible for 144 hour visa free transit in Guangdong province, you have to fly between the two cities on direct flight.

Alternatively, you can construct your itinerary to look like:

Japan - Guangzhou - Hong Kong.

In this case, you are allowed to take a conventional train from Guangzhou East to Hong Kong Hung Hom railway station and also a ferries from both Nansha (south from Guangzhou) and from Shenzhen Shekou terminal. Please, keep in mind that you will have to book a ticket for a train/ferry in advance if you decide to use this itinerary for your 144 hour visa free transit in Guangdong province.

By the way, if you are a Japanese citizen, you can travel on your mentioned itinerary by every mode of transport because Japanese citizens do not need a visa for up to 15 days stay in China.
