It is possible to benefit from the 144h scale in Shanghai if I do:

Osaka - Seoul
Seoul - Shanghai
Shanghai - Osaka?

If so, do I need to leave the airport in Seoul or having arrived in Shanghai from Seoul and showing my flight back to Osaka is enough?

Yes, you are eligible for 144 hours visa free policy. In addition, it will be ok for you to show the flight back to Osaka.
