About:24-Hour Visa-Free Transit

I am an India passport holder travelling from Canada to India via two stopover in China

First stopover at Nanking at 14.45 hrs with a layover time of 3.30 hrs.
Next flight leaving for Kunming at 18.00 hrs. Arriving at kunming at 21.00 hrs.
Layover at Kunming for 2 hrs.
Next flight to India from Kunming at 23.30 hrs.
All arrival and departure in/from China on the same day.
Airline: China Eastern Airlines
Do I require to take the 24 hr. Permit or an stay at the international transit area.

Your are definitely eligible for 24 hour visa free transit during your entire transit in China. In other words, you don't need a visa of any kind. You will declare at check-in in Canada that you will travel to India via China using 24 hour visa free transit. You should have with you your flight tickets in hard copy (e-ticket printouts) that you will present at check-in. After a short inspection, you will be allowed to board the plane.

Upon your landing in Nanjing, you will pass the immigration inspection and you will also apply for 24 hour free entry permit that will be definitely granted to you because you will continue your trip in domestic segment of your flight. You will be allowed to exit the airport and to explore the cities where you will have your layovers, but since your layovers are very, very short, you will have to accomplish all your formalities to catch your following flight.
