About:240-Hour Visa-Free Transit

Hi, if we travel from HK to Guangzhou then to Macau back to HK, are we qualified for 144 hr exemp ?

We will be travelling via coach from HK to Guangzhou and back to Macau via coach/bus

No. You need a visa. Only arrivals by air are eligible for 144 hours transit without visa.
