About:72-Hour Visa-Free Transit

Can an American take advantage of this twice in 30 days?

If it works, I hope to travel from Europe to Chinese City A, onward to S Korea, Then S Korea to Chinese City B, onward to USA.

For instance:

Barcelona --> Beijing (72 hr stay) --> Seoul
I will stay in Korea for about a week
Seoul --> Shanghai (72 hr stay) --> USA

All airline tickets will be bought in advance.

Yes, of course. There is no limit as to how often one can use visa free transit scheme in China.

Your itinerary is definitely eligible, but you should take care that your flights to/from Beijing and to/from Shanghai, don't have any additional landing within mainland China. Also, your flights from Barcelona to Beijing and Shanghai to US should not have a landing in South Korea.

One more detail. In reality, you are eligible for 144 and not for 72 hour visa free transit in Beijing/Shanghai because 144 hours has replaced 72 hours a couple of years ago.
