Do I need visa to leave Hong Kong hotel for overnight stay before traveling on?

Hold USA passport. Arrive in Hong Kong with 12 hour layover before continuing on to Vietnam. Do I need a transit visa to leave the airport for an overnight hotel stay?

No, you don't. As US citizen you are allowed to exit the airport and to stay overnight in hotel because US citizens don't require any kind of visa for Hong Kong.
