Seoul to Shanghai to Los Angeles

I would be arriving from Seoul on a Thursday afternoon, and flying out of Shanghai that Saturday morning. However, my flight has a layover in Guangzhou for almost 12 hours before returning back to LA.

I believe my initial trip to Shanghai is covered in the 72 hour visa free eligibility. But with that layover in Guangzhou do I need to apply for a visa to step out of the airport and look around before flying back to LA?

Your itinerary definitely requires a visa that you should obtain in advance. You are not eligible for 144 hours visa free transit because you will travel consecutively to two different visa regions which is not allowed and you are also not eligible for 24 hours visa free transit that allows more than one stop in China, but has a limit of only 24 hours, while your itinerary takes more than that.
