If I am flying HKG-PVG-ORD, can I do the 72-hour layover and take the high speed train to Beijing and back if within the 72 hours?

No, you cannot do that. If you want to visit Beijing during the layover in Shanghai, you cannot use the free-transit. You should apply for a visa for the trip.
