About:72-Hour Visa-Free Transit

Shanghai to HK to Shenzhen

If I land in Shanghai for 1 night then fly to HK and take the ferry to Shenzhen will I require 2 x 72 hour visas.

Assuming that you are not arriving at Shanghai from HK, you can use 144-hr transit in Shanghai. For Shenzhen, however, you need to pay for a 5-day visa on arrival.
