About:72-Hour Visa-Free Transit

On the departure destination, is it the final destination that counts?

I am traveling from the US to Japan, will stay for over 24 hours, and then continue to Beijing where I will stay for 35 hours. The return ticket to the US is with a 2 hour layover in Japan, so for transit entry permit purposes, will the exit destination be considered the two hour stopover point ( Japan) or the final destination (US)?

You need a visa. Chinese immigration will consider your trip as Japan-China-Japan, which is not a transit.
