I will land on PVG but leave from HGH.

I am an American and am going to attend an academic conference in Hangzhou.

I will arrive at Shanghai Pudong Airport from Newark, USA (EWR) on Friday noon. Then take a long-distance bus to Hangzhou City. Then attending the conference on Saturday and Sunday. The only place to go is around Hangzhou City and riding a bus from PVG to Hangzhou. On Monday I will take a flight from Hangzhou (HGH) to San Francisco (SFO), not PVG to SFO.

Can I still use the 144hr transit visa, even though leaving from HGH not from PVG?

It is no problem to leave from Hangzhou Airport, but since you will fly back to USA, I am afraid you are not qualified for 144-hour visa free transit policy. If you want to enjoy the policy, you may consider going to Shanghai from US, transfer to Hangzhou, fly from Hangzhou to Hongkong which is regarded as the third region, and then fly back to USA.
