can I travel Sydney - Guangzhou - (flight transit in Shanghai) to Singpore on the 72 hour visa?

No, you cannot enjoy the 72h visa free policy for u will transit in two cities. However, if your total transit duration is within 24h in the two cities, just apply for a stay permit in Shanghai and you don't need a visa then.1
