You obviously plan to travel:
US - Hong Kong - Shenzhen (mainland China) - Macau - Hong Kong - Tokyo
If this is the case, your itinerary is definitely eligible for 240 hour visa free transit and you do not need any kind of visa. My recommendation for you is to take a train from Hong Kong West Kowloon station to Futian station in Shenzhen. As far as your exit from Shenzhen to Macau is concerned, you will have to buy your Shekou (a port in Shenzhen) to Macau ferry ticket while still in Taiwan or Hong Kong. You will have to enter mainland China with a ticket at hand that will guarantee that you will exit toward the third country/eligible territory.
It really does not matter how long (short) you will stay in Macau and your use of Macau is a very creative and acceptable way to travel. Have a nice visit to China.
