No, you cannot do that. The basic rule of 240 hour visa free transit clearly requires that the immediate points before and after mainland China have to be in two different countries or eligible territories (eligible territories are Hong Kong, Taiwan and Macau) and your itinerary has both of its immediate points in just one eligible territory - Hong Kong.
The solution for your itinerary is to insert nearby Macau into the equation. You can take a ferry from Hong Kong to Macau and as soon as you have arrived there, you will take a ferry from Macau to Shekou in Shenzhen. From Shenzhen you can proceed to other parts of mainland China, Guangzhou included. After your stay in mainland China you will exit it toward Hong Kong. Like this you will have eligible itinerary: Macau - mainland China - Hong Kong.
Please, take care that you will have to buy your train exit ticket from mainland China to Hong Kong before you depart toward mainland China because you will be asked to present it in both Macau and upon your arrival in Shekou port in Shenzhen. One more detail, when you arrive in Shenzhen do not apply for 5 day Shenzhen VOA because this is not for you because you will look for a temporary entry permit / 240 hour visa free transit.
