Tom, you need a visa as a US citizen, but your wife needs a Mainland Travel Permit for Hong Kong and Macao Residents usually called Home Return Permit. Hopefully she has one.
When you arrive at Lo Wu you will clear Hong Kong exit formalities. Lo Wu is so easily reachable by a MTR suburban train from the center of Hong Kong. After that you will shortly walk until you reach Luohu that is a mainland Chinese counterpart of Hong Kong run Lo Wu. Here you can both walk to the visa office, but only you will apply while your wife will wait for you. When you receive a Shenzhen 5 day VOA, you will both head to the mainland Chinese immigration line and cross into mainland China. In other words, you do not have to separate one from each other at all.
The price is 130 CNY (RMB) and you can pay it in cash or by your bank card (practically all overseas bank cards, debit and credit are accepted). If you decide to pay by cash and you do not have exact amount, a visa office employee will return a change in Chinese cash, of course. When you are over with your visit to Shenzhen, you can return to Hong Kong by taking any border crossing and any mean of transport, high speed train included. Have a nice visit to Shenzhen.
