If you are a citizen of Singapore, you do not have to bother about the formalities related to 24 or 240 hour visa free transit since the citizens of Singapore can enter mainland China (Shenzhen included) absolutely visa free for up to 30 days. In other words, it is up to you how will you enter and also how will you exit mainland China, free from 24/240 hour visa free limitations.
If you are not a citizen of Singapore, but if you are a citizen of 55 countries that are eligible for 240 hour visa free transit a route from south Asia to Shenzhen and after 7 days to Hong Kong is acceptable and eligible for 240 hour visa free transit. Also, if you can buy and have at hand your bus ticket from Shenzhen to Hong Kong that should be fine to satisfy the rules of 240 hour visa free transit. You are warned that you will have to buy your ticket in advance, even before you depart from south Asia toward Shenzhen because you will be asked to present it in both the country that you will use to depart to Shenzhen from and also upon your landing in Shenzhen. Also, it is important to understand that it has to be a single, through ticket and not a segmented one.
If I may, I would suggest you not to bother with the bus ticket because it is not usually (typically) used for 240 hour visa free transit purposes, but to book a train ticket from Shenzhen (preferably Futian station if you plan to stay in the central area of Shenzhen) to Hong Kong West Kowloon station. This is much easier, more straightforward and so well know among the check-in and immigration authorities. Your entire journey will be much better with an exit train ticket from Shenzhen to Hong Kong compared to bus ticket. In any case, I leave a final decision to you.
