You can do that, but under two conditions:
1. You have to arrive into Guangzhou by direct flight from a country that is different from both mainland China and Hong Kong and you are not allowed to have any additional landing within mainland China and Hong Kong before Guangzhou.
2. You cannot take a train from Guangzhou to Hong Kong. This was possible several years ago when conventional trains (slow one) were still in use. With the arrival of high speed trains and termination of conventional ones, this is not possible any more. You are advised to take a ferry from Nansha port located south from Guangzhou and reachable by metro directly to Hong Kong. In order to do that, you will have to buy your ferry ticket in advance, even before you depart for Guangzhou, through CKS - Chu Kong Passenger Transport company, and you will have to print it out and have that printout with you at hand. Also, you will really have to take exactly that ferry you have bought your ticket for.
If you do like this, you are free to enjoy 144 hour visa free transit in the entire Guangdong province and you do not need any kind of visa.
