About:144-Hour Visa-Free Transit

Fly from Osaka to Guangzhou (transfer n layover 3 hour in Hangzhou). Train to Hong Kong 4 days later

Will fly from Osaka to Guangzhou - flight has a transfer and 3 hour layover in Hangzhou. Stay in Guangzhou for 4 days and take train to Hong Kong. Do I qualify for 144 visa free transit?

This itinerary does not qualify for 144 hours visa-free transit. You have stops in two different parts of China (Hangzhou, Guangzhou) and the train to Hong Kong is no longer an option. So you will need a Chinese visa in advance.
