Your itinerary is definitely eligible for 144 hour visa free transit and this is valid for entire Guangdong province.

The best way to exit Guangdong province toward Hong Kong is by taking a ferry from:
- Nansha (located south from Guanzhou)
- Shekou (located in Shenzhen)

Both lines are performed by CKS - Chu Kong Passenger Transport.

You will have to buy and print out the ticket for the ferry and have it with you in hard copy. Also, you will have to take exactly that ferry you have bought your ticket for before you have departed from Singapore.

You are informed that there is no eligible train option any more. The older, conventional through train is discontinued because it is slow and high speed trains are not eligible.

You can also think about the flight from Guangzhou to Hong Kong, although I consider ferry to be the best option.
