Part 1:

Actually, this is not possible because you cannot move within two different transit areas. In your case, you have Guangdong (Guangzhou and Shenzhen) as one and Xiamen as another transit area.

So, you should be a bit more creative. For example, your itinerary can look something like this:

Singapore - Hong Kong - Macau - Xiamen (144 h visa free transit) - Hong Kong

This is the first segment and as you can see, the immediate points in before and after your mainland Chinese destination are located in two different geographical entities. Now, the segment with Guangdong province as a continuation of your entire itinerary:

Hong Kong - Guangzhou (by air only) - Macau (by ferry from Shenzhen)

In order to be eligible for this, visa free transit scheme, you will have to fly from Hong Kong to Guangzhou and then take a ferry from Shekou port in Shenzhen to Macau. The reason is simple... The only eligible way to enjoy 144 hour visa free transit scheme in entire Guangdong province is to arrive by air.

If you decide to use this option, you will have to buy also your ferry ticket in advance, print it out and have it with you. Also, you will really have to take exactly that ferry you have bought your ticket for before your departure toward mainland China.

If you need an additional day in Shenzhen after you have arrived in Macau, you can take a ferry from Macau to Shenzhen and apply there for 5 days Shenzhen visa on arrival. It costs just 168 CNY and after your stay in Shenzhen you can exit to any direction and by any mean of transport, including a possible direct, nonstop flights from Shenzhen to Singapore. You can, when using this visa on arrival arrive in Hong Kong and Macau too... As you wish.

It is just important you do not confuse 144 hour visa free transit and this 5 days Shenzhen visa on arrival. These are totally different things, but in your case both can prove to be useful.
