In my opinion, if you intend to visit Shenzhen only, the best option is to apply for a 5 days Shenzhen visa on arrival at Luohu (Lo Wu) or Huanggang border crossings. It is quite cheap for Singaporean citizens and requires minimal formalities. After your stay in Shenzhen, you can return to Hong Kong by any mean of transport, high speed train included and you can also use any border crossing.
If you intend to visit the wider area or to be precise Guangdong province, you should take a plane from Hong Kong to Guangzhou and after your stay there exit by ferry to Macau. This will allow you 144 hour visa free transit within the entire Guangdong province. Please, keep in mind that you will have to buy a ferry ticket in advance, print it out and have a hard copy with you as a proof of your transit.
