About:240-Hour Visa-Free Transit

Is 144-hour visa possible in this case?

I am a US citizen living in Japan. I want to fly to Seoul and stay there for one day. From Seoul, I plan to fly to Beijing and stay there Beijing -> Japan), is the 144-hour layover visa valid in this case? Thank you.

My question was cut off... please read this instead:

I am a US citizen living in Japan. I want to fly to Seoul and stay there for one day. From Seoul, I plan to fly to Beijing and stay there for 5 days, then return to Japan -- but on my return flight to Japan I have a layover at Seoul again. As long as I don't leave the airport/immigration in Seoul on the way home, because I am flying "Seoul to Beijing to Japan", is the 144-hour layover visa valid in this case? Thank you.

Your ticket will be Japan-Seoul-Beijing-Seoul-Japan therefore transit free visa will not apply.

Where you stop immediately before and immediately after determine eligibility. Seoul > China > Seoul is not a transit. It doesn't matter where the ultimate destination is.
