About:72-Hour Visa-Free Transit

Follow up on Japan to Vietnam

Sorry for the two part question! Just realized that on the return flight there would also be a 21 hour layover in Xiamen. Would I be able to leave the airport again? It would be within ten days.

Sum up
Flight from Japan to Vietnam - 25 hour layover in Vietnam (eligible for the 72 hour visa)
Flight from Vietnam back to Japan - 21 hour layover.

Alex, no need to worry! You can use the 24 hour direct transit when you return. You can leave the airport again as long as you apply for a temporary stay permit which is free of charge at the airport.
