About:144-Hour Visa-Free Transit

USA to China with connection in Japan

I plan to fly from the US to Shanghai with a connection in Japan. I will
then return to Japan for a week before returning to the US.

Do I still qualify for the 144 hour free transit with that 2 hour connection in Japan
outbound?

Of course not. Japan-Shanghai-Japan isn't a transit. It doesn't matter how long you are in Japan for. You either need a visa or you need to change your itinerary.
