Can I come to Shanghai without a visa for the 144 hours if I'm flying San Francisco to Shanghai to Tokyo ? Can I fly standby? Or would I need to purchase a ticket from Shanghai to Tokyo? Thanks

The travel route qualifies for the 144 hour free transit, but you cannot use it if you fly standby. You need to purchase tickets for this journey.
