About:72-Hour Visa-Free Transit

I am from Russia and currently in Hong Kong. I would like to go to Guangzhou for a day.

I am leaving to Russia 3 days later. Can I use a direct transit visa that would be Hong Kong - Guangzhou - Moscow (via Hong Kong)?

No, because your route is Hong Kong - Guangzhou - Hong Kong you're not in transit. You need a visa for that.
