If I am a US citizen and wanted to do a weekend in Shanghai from Taipei, do I need a Visa?

Yes you need it if you do a roundtrip from Taiwan.If you fly to a 3rd country you can use the 144h twov in Shanghai but if you fly back to the country you came from you´re not qualified for the free transit programm.
