About:72-Hour Visa-Free Transit

I am a USA citizen working in hong kong. Am I able to use the 72 hours regardless of how I enter?

I am looking at various options to enter China from Hong Kong. Will I receive the 72 hours regardless of how I enter mainland PRC?
