I am a United States Citizen and traveling to Thailand but will have a 16 hour layover in Guangzhou.

May I leave the airport and travel to Hong Kong and then get back on the plane and go to Phuket, Thailand with the 72 hour visa?

The answer is NO.
As you only layover for 16h, the 72h visa free policy is not needed. You are eligible for the 24h visa free transit. Upon landing, just apply for a stay permit and you will be allowed to leave the airport. However, you cannot go to HK; with such a transit, you can only stay in GZ.
