Hi, me and my friends are going to Macau this weekend for just a day tour.

can you please help me have the best itinerary for a full day tour for a group of 6, and less expenses.

Well, according to your limited time, you need to make full use of time.
The suggested itinerary is like the following:
Ruins of St. Paul's, Senado Square, Macao Tower, A-Ma Temple, Fisherman's Wharf, Grand Prix Museum, Wine Museum

Hi Adele,
Thank you very much for this! Are all of this places cheap or not that expensive? We are converting 500HKD only for Macao, as we don't have that much budget.. Can you please help us also which one of these should we go first so we can do all this in a day(what about The Venetian is it possible to include that?).. also i was informed that there are free bus rides around Macao, how can we avail those and what are those buses? please help!

I don't agree with Adele. That is too much for one day tour. Moreover, I am hoping you changed 500HKD each person, because only the single ride on ferry costs 160-170HKD per person. With the rest budget, you can still have a good trip there. Here is my suggested itinerary:
Upon getting off ferry at Maritime Ferry Terminal, walk to Fisherman Wharf (free) for a visit. Then head to the Wine Museum (15MOP) and Grand Prix Musuem (10MOP). Take bus no. 3 or 10 (I remember it costs 3-4MOP) to Avenida de Almeida Ribeiro and walk to Senado Square and the Ruins for a visit. Thereafter, walk to Hotel Sintra to take the free shuttle bus to City of Dreams, which is right oppsite the Venetian. Only visiting the Venetian is for free~

I forget to mention that HKD can widely be used in Macao.
