Hey, the suggested route would be like this:
1. Grand Lisboa - Senado Square – Ruins of St. Paul – Macao Museum – Guia Fortress. For these attractions, you can just go by walk follow the sequence I listed.
2. From Guia Fortress to Fisherman’s Wharf: you can walk northwards to Two Dragonmaw Park to take bus no. 23 there, and get off at Tourism Activity Center. And then walk eastwards to reach it.
3. For the others from the Fisnerman’s Wharf: you can take bus no. 3A, 10A, or 12 and get off at Madrid Street. Walk southeast you can reach the Kun Lam Statue. After then, you can walk to the northwest for about 1200 yards to reach the Wynn Hotel. And then take bus no. 9 to reach Macao Tower.

Hope helpful.
