Which kind of transportation method do you need?

I think the flight is the best method. You can fly from Kashgar to Urumqi and the fly to Haikou.
The flight from Kashgar to Urumqi takes around 100 minutes. From Urumqi to your destination city takes around 5 hours, because of a stopover in Chongqing or Chengdu.

How can I travel by train for the trip? And how much does it cost for both methods?

I'm afraid there is no direct train service for the journey. Then you need take a train to Urumqi and then transfer at Xian to catch a train to Haikou. It shall take much longer. As for the timetable and fare, you can search on the train page at this website.

As the air ticket fare also varies from time to time, it is hard to tell you the exact fee, especially without knowing your travels date. Just search at this website also.
