Sc, unfortunately, your itinerary is not eligible for 240 hour visa free transit because your trip is not a transit, but just a round trip from Hong Kong. The basic rule of 240 hour visa free transit states that the immediate points before and after mainland China have to be in two different countries or eligible territories (eligible territories are Hong Kong, Taiwan and Macau). As you can see, your booth immediate points are in one territory - Hong Kong. However, I would like to suggest you one relatively easy and creative solution for your needs. You can depart toward mainland China from Macau by ferry to Shekou in Shenzhen. This route is eligible way how to enter mainland China from Macau. So, when you are in Shenzhen, you can take a domestic flight from Shenzhen to several destinations in Yunnan (Kunming, Lijiang, Dali...). In other words, you can explore Yunnan freely, on your own. After your stay in Yunnan province, you can take a direct flight from Kunming to Hong Kong or even better (because of wider choice), from some of Yunnan airports back to Shenzhen and from Shenzhen by a high speed train to Hong Kong. So, if you decide to take this option, you should just have in mind that you will need your Shenzhen to Hong Kong train ticket already bought, even before you board the ferry from Macau to Shekou (Shenzhen). In other words, like this you will have an itinerary Macau - mainland China - Hong Kong and that is an eligible itinerary.
