Unfortunately, your planned itinerary is not eligible for any kind of visa free transit and you need a visa obtained in advance in order to perform your itinerary.
Simply speaking, the basic rule of 144 hour visa free transit clearly states that you are allowed to have just one landing in mainland China and that the immediate point before and after your mainland Chinese destination have to be in two different countries or eligible territories (eligible territories are Hong Kong, Taiwan and Macau). As you can see, you have two mainland Chinese destinations in a row and the situation of this kind immediately disqualifies you from using this visa free transit scheme.
In order to be eligible for 144 hour visa free transit you will have to be creative and to use, for example Taiwan in order to make your itinerary eligible. For example:
Singapore - Shanghai - Taiwan - Guangzhou - Hong Kong
Itinerary of this kind is eligible for 144 hour visa free transit in both Shanghai and Guangzhou because the immediate are in both cases located in two different countries/eligible territories.
You can exit from Guangzhou to Hong Kong by taking a high speed train. You cannot use a car because you will have to produce a ticket before your arrival in Guangzhou to prove that you will be really in a transit.
In the end, I encourage you to use correct terminology because it is not 144 hours visa, but 144 hour visa free transit, so there is no visa of any kind.
